The Church of Santa Maria delle Vergini was dedicated to Santa Rita in 1904, when it was given to the Brotherhood of the Holy Thorn of Our Lord Jesus Christ and St. Rita of Cascia (founded in 1655), previously in the Church of St. Rita (once San Biagio del Mercato) demolished to make way for the Monument to Victor Emanuel IInd.

The Monastery of St. Mary of the Virgins was set up by Father Pompeo Pateri of the Congregation of the Oratory of St. Philip Neri, who in 1593 had founded the Collegio della SS. Madonna del Rifugio on the Quirinal Hill to provide hospital care for poor unmarried women and orphans. At a later date, the building that housed this charitable institute was knocked

down to make way for a new home for Cardinal Scipione Borghese (later Palazzo Pallavicini-Rospigliosi), and in 1612, Pateri obtained permission from Pope Paul Vth to move the "Collegio" into new premises in Palazzo Taverna, in front of the "Monastero dell'Umiltà". In 1613 the Institute (then run according to St. Augustin's doctrine) was turned into a convent for seclusion nuns and dedicated to Santa Maria delle Vergini Assunta in Cielo though it still carried on taking in and caring for poor spinsters according to its founder's wishes. In 1615, a church was built next to the monastery but this soon proved to be too small for the needs of the growing community, so the nuns appointed Architect Francesco Peparelli to rebuild it (1634-1636). Finally in 1656, under Gian Lorenzo Bernini's guidance with Domenico Castelli as works supervisor, foundations were laid for a large new monastery that was completed in 1660 (partly demolished some time around 1950).

The church was later embellished with a new main altar executed by Matthia de' Rossi (1681) and the façade design with two rows of half columns and a large central window is attributed to the same artist (1696-97); the bell tower was added on in 1689.

The interior is of Greek cross form and still contains the original sumptuously Baroque decorations: the exquisite stuccoes were carved by Filippo Carcani and Ludovico

Gimignani painted a fresco of the Holy Trinity under the arch of the main chape; an Assumption of the Virgin was painted in 1681 to replace the main altarpiece of the same subject by Andrea Camassei, and is now in the Church of St. Pudenziana. The fresco in the dome depicting The Glory of Heaven is attributed to Michelangelo Ricciolini (1695 approximately). Also remarkable is a monumental wooden choir with carvings and guilding in the wall of the counter-façade.
